An IL-1 antibody (interleukin-1 antibody) is a class of monoclonal antibodies designed to target and neutralize interleukin-1 (IL-1) signaling, a key pathway in the inflammatory response. The IL-1 superfamily consists of major pro-inflammatory cytokines, primarily IL-1α and IL-1β, which bind to the type 1 IL-1 receptor (IL-1R1) to recruit the IL-1 receptor accessory protein (IL-1RAcP) and initiate downstream inflammatory cascades via NF-κB and MAPK pathways. Monoclonal antibodies in this class can specifically target IL-1α (e.g., bermekimab), IL-1β (e.g., canakinumab, gevokizumab, abdakibart), or both (e.g., lutikizumab). These therapeutics are developed and approved for a wide range of autoinflammatory, rheumatologic, cardiovascular, and dermatological conditions, such as cryopyrin-associated periodic syndromes (CAPS), systemic juvenile idiopathic arthritis, gouty arthritis, hidradenitis suppurativa, and cardiovascular risk reduction.
