An IL-10RA blocking antibody is a monoclonal antibody designed to bind to the Interleukin-10 receptor subunit alpha (IL-10RA), thereby inhibiting the signaling of the immunosuppressive cytokine IL-10. In the context of oncology, particularly in primary cutaneous diffuse large B-cell lymphoma (PCDLBCL-LT), IL-10 secreted by tumor cells induces an immunosuppressive M2-like phenotype in tumor-associated macrophages (TAMs). By blocking the IL-10/IL-10RA axis, these antibodies can reverse macrophage-mediated immunosuppression, enhance T-cell activity, and potentially sensitize tumor cells to chemotherapy. Research presented at ASH 2025 indicates that IL-10RA blockade can fully reverse the phenotypic changes induced by PCDLBCL-LT cells on macrophages, suggesting a potential therapeutic vulnerability in this disease.
