Overview

IL-12 gene-transfected KB cells are genetically engineered KB carcinoma cells (a human epithelial cell line derived from a nasopharyngeal carcinoma) that have been modified to express and secrete interleukin-12 (IL-12). These cells are designed as a form of cancer vaccine: they secrete IL-12 locally when administered, stimulating a local and systemic immune response against tumor cells. The mechanism relies on IL-12's potent activation of natural killer (NK) cells and cytotoxic T lymphocytes, along with inducing interferon-gamma (IFN-γ) production, thereby enhancing antitumor immunity. The principal documented indication is as an investigational therapy or immunotherapy vaccine for head and neck carcinoma and potentially other tumors. Preclinical data (mouse models) showed that vaccination with IL-12 gene-transfected KB cells can significantly inhibit tumor growth, especially when combined with cells secreting other cytokines such as IL-2[5].
