IL-13Rα2 ε-TRuC is an experimental T-cell receptor fusion construct (TRuC) therapy developed by TCR2 Therapeutics for the treatment of glioblastoma multiforme (GBM) and other solid tumors. The therapy consists of autologous T cells engineered to express a fusion protein comprising a single-domain antibody (VHH) specific for the tumor-associated antigen Interleukin-13 receptor alpha 2 (IL-13Rα2) linked to the extracellular N-terminus of the CD3ε subunit of the T-cell receptor (TCR) complex. Unlike traditional Chimeric Antigen Receptor (CAR) T-cell therapies that utilize isolated CD3ζ signaling domains, TRuCs are designed to integrate into the endogenous TCR complex, thereby harnessing the full physiological signaling machinery of the T cell. This approach is intended to provide more physiological T-cell activation, potentially leading to improved tumor penetration and efficacy in solid tumors with a more controlled cytokine release profile.
