Overview

IL-13Rα2 CAR-T cells are an autologous chimeric antigen receptor (CAR) T-cell therapy designed to target interleukin-13 receptor alpha 2 (IL-13Rα2), a cell surface receptor that is highly overexpressed in the majority of high-grade gliomas, including glioblastoma multiforme (GBM), while remaining absent or minimally expressed in normal brain tissue. The CAR construct typically utilizes a membrane-tethered IL-13 cytokine ligand, often containing a specific mutation (e.g., E13Y) to increase binding affinity for IL-13Rα2 and reduce interaction with the more ubiquitous IL-13Rα1. Upon administration, these engineered T cells recognize and bind to IL-13Rα2-positive tumor cells, triggering T-cell activation, cytokine release, and direct cytotoxic killing of the glioma cells. This specific therapeutic candidate is being investigated by The First Affiliated Hospital of Suzhou University in Phase I clinical trials for patients with advanced or recurrent high-grade gliomas.
