Overview

IL-13Rα2 DNA vaccine is an experimental DNA vaccine designed to encode the interleukin-13 receptor alpha 2 (IL-13Rα2), a protein aberrantly overexpressed in multiple human cancers, including glioblastoma, breast, pancreatic, kidney, ovarian, and sarcoma[1][2][3]. The vaccine aims to elicit a targeted immune response—particularly T cell-mediated cytotoxicity—against tumor cells expressing IL-13Rα2, thereby inhibiting tumor growth and metastasis[1][2]. The mechanism of action includes induction of IL-13Rα2-specific cytotoxic T lymphocytes and antibody responses, potentially enhanced when used in a prime-boost protocol (e.g., DNA vaccine priming followed by protein/adjuvant boosting)[1][2][3].\n\nIn preclinical murine models, IL-13Rα2 DNA vaccination has shown significant anti-tumor effects both prophylactically and therapeutically, resulting in reduced tumor growth, decreased metastases, and prolonged survival[1][2][3]. It is being explored primarily for treatment of IL-13Rα2-positive malignancies and as a combination partner with IL-13Rα2-targeted cytotoxins or other immunotherapies[1][2]. As of 2025, there are no reports of clinical trials in humans with this specific DNA vaccine, though peptide/antigen-pulsed dendritic cell vaccines using IL-13Rα2 have advanced to clinical evaluation[2].
