Overview

IL-15 activated allogeneic NK cells is an investigational adoptive cell therapy developed by Leiden University Medical Center (LUMC). The therapy involves natural killer (NK) cells harvested from an allogeneic donor, typically the same individual who provided stem cells for a prior hematopoietic stem cell transplant. These NK cells are activated ex vivo using interleukin-15 (IL-15), a cytokine that significantly enhances their survival, proliferation, and cytotoxic potential. Once infused into the patient, these activated NK cells target and eliminate malignant leukemic blasts by recognizing the absence of self-MHC class I molecules and through various germline-encoded activating receptors. The therapy is currently being evaluated in feasibility studies for pediatric patients with relapsed or refractory leukemia following allogeneic stem cell transplantation to determine its safety and potential efficacy in preventing or treating relapse.
