IL-15-Fc is a recombinant fusion protein designed to harness the immunostimulatory properties of interleukin-15 (IL-15) while improving its therapeutic profile. It typically consists of the IL-15 cytokine fused to the Fc region of a human immunoglobulin G (IgG), which extends the half-life of the cytokine through increased molecular size and neonatal Fc receptor (FcRn)-mediated recycling. IL-15 is a critical factor for the survival, proliferation, and activation of natural killer (NK) cells and effector CD8+ T cells. Unlike IL-2, IL-15 does not primarily stimulate regulatory T cells (Tregs) or induce vascular leak syndrome at therapeutic doses, making it a preferred candidate for oncology and infectious disease applications. The IL-15-Fc platform includes various configurations, such as direct fusions or complexes involving the IL-15 receptor alpha (IL-15Rα) sushi domain, all aimed at enhancing the potency and trans-presentation of IL-15 to its signaling receptor complex (CD122 and CD132).
