Drug intelligence / Profile preview
An IL-15 neutralizing antibody is a therapeutic monoclonal antibody designed to bind to and neutralize interleukin-15 (IL-15), a pleiotropic cytokine that plays a critical role in the development, survival, and activation of T cells (particularly CD8+ memory T cells) and natural killer (NK) cells. Dysregulation and overexpression of IL-15 are implicated in various autoimmune and inflammatory disorders, such as celiac disease, vitiligo, alopecia areata, and rheumatoid arthritis, where it drives tissue destruction and chronic inflammation. By blocking IL-15 signaling, these neutralizing antibodies inhibit the activation of cytotoxic lymphocytes and downstream inflammatory cascades, offering a targeted therapeutic approach for IL-15-mediated diseases. Several clinical-stage candidates, including TEV-408, CALY-002, and ordesekimab (AMG 714/PRV-015), are being evaluated in clinical trials for indications like celiac disease and vitiligo.
