Overview

IL-15-stimulated allogeneic NK cells is an investigational adoptive cell therapy consisting of donor-derived natural killer (NK) cells (CD56+, CD3-) isolated from a haploidentical allogeneic donor. The production process involves a double immunomagnetic depletion and enrichment protocol to isolate the NK cells, followed by ex vivo stimulation with interleukin-15 (IL-15) for approximately 12 to 20 hours. This stimulation is intended to enhance the anti-leukemic activity, survival, and cytotoxicity of the NK cells. The therapy is primarily being evaluated for the treatment of high-risk acute myeloid leukemia (AML) in adult patients undergoing unmanipulated haploidentical allogeneic stem cell transplantation with post-transplant cyclophosphamide. The mechanism leverages the graft-versus-leukemia (GvL) effect, where the activated NK cells target and eliminate residual leukemic cells during immune reconstitution.
