Overview

The **IL-15-transpresenting WT1-targeted autologous dendritic cell vaccine** (also known as the **IL15/IL15Ra/WT1 DC vaccine**) is an investigational, personalized cell-based immunotherapy developed by Antwerp University Hospital. It consists of autologous monocyte-derived dendritic cells engineered via electroporation with messenger RNA (mRNA) encoding both interleukin-15 (IL-15) and interleukin-15 receptor alpha (IL-15Rα) to transpresent IL-15 on their cell surface, alongside presentation of the Wilms' Tumor Protein 1 (WT1) antigen. This dual mechanism is designed to enhance the immunogenicity of the dendritic cells, boosting natural killer (NK) and CD8+ T cell activation and survival while directing a cytotoxic immune response specifically against WT1-expressing cancer cells. The vaccine is currently being evaluated in a Phase 1 first-in-human clinical trial (NCT05964361) for patients with advanced or refractory solid tumors of the pancreas, esophagus, liver, and ovaries.
