IL-1RAP CAR NK cells are an investigational cell therapy consisting of natural killer (NK) cells engineered to express a chimeric antigen receptor (CAR) targeting the interleukin-1 receptor accessory protein (IL-1RAP). IL-1RAP is a co-receptor for IL-1, IL-33, and IL-36, and is highly expressed on acute myeloid leukemia (AML) cells and leukemic stem cells, while being absent on normal hematopoietic stem cells, making it a promising target for immunotherapy. These CAR NK cells are designed to induce targeted cytotoxicity against AML cells. Research led by Mitchell S. Cairo and colleagues has explored enhancing the activity of these cells using IL-15 agonists like NKTR-255 or tri-specific killer cell engagers (TriKEs) such as cam161533 to improve persistence and potency.
