IL-2 gene-transduced allogeneic neuroblastoma cells is an experimental cell-based immunotherapy developed by St. Jude Children's Research Hospital for the treatment of relapsed or refractory neuroblastoma. The therapy consists of allogeneic (donor-derived) neuroblastoma tumor cells that have been genetically modified using retroviral gene transfer to express and secrete human interleukin-2 (IL-2). Functioning as a cancer vaccine, the IL-2-secreting tumor cells are designed to stimulate a potent anti-tumor immune response, particularly by enhancing the activity of cytotoxic T lymphocytes and natural killer cells against neuroblastoma-specific antigens. This biologic therapy was evaluated in Phase I clinical trials where it was administered via subcutaneous injection to induce a local, polyclonal T-cell infiltrate and systemic anti-tumor immunity.
