IL-2 pre-activated mismatched T + NK + NKT cells is a cell therapy product composed of a mixture of allogeneic (mismatched) T cells, natural killer (NK) cells, and natural killer T (NKT) cells that have been activated ex vivo with interleukin-2 (IL-2). This combination leverages the cytotoxic potential of multiple immune effector cell types to target cancer or other pathological conditions. The use of IL-2 in the activation process enhances the cytotoxicity and proliferation capacity of these lymphocytes, particularly boosting NK cell reactivity against tumor targets[1][3][5]. The "mismatched" aspect refers to partial HLA incompatibility between donor and recipient, which can increase anti-tumor activity but also requires careful management to avoid graft-versus-host disease. This approach is under investigation primarily for hematologic malignancies such as drug-resistant leukemias[1], with some studies reporting partial anti-tumor responses in solid tumors as well[5]. The mechanism involves direct cytolytic activity against malignant or infected host cells through both innate and adaptive immune pathways.
