IL-2-transfected CIK cells are an autologous cell therapy consisting of cytokine-induced killer (CIK) cells—a heterogeneous population of T lymphocytes with natural killer (NK) cell-like activity—genetically modified to express interleukin-2 (IL-2). CIK cells are typically generated from peripheral blood mononuclear cells (PBMCs) through ex vivo stimulation with IFN-gamma, anti-CD3 antibodies, and IL-2. In this specific formulation, the cells are transfected (often via electroporation with a plasmid) with the IL-2 gene to provide a localized, autocrine source of the cytokine, thereby enhancing the cells' proliferation, survival, and cytotoxic activity against tumor cells without the systemic toxicity associated with high-dose IL-2 administration. This approach has been investigated in Phase I trials for metastatic renal cell carcinoma, colorectal carcinoma, and lymphoma.
