Overview

IL-2-transfected CIK cells are an autologous adoptive cell therapy developed by researchers at the Humboldt University of Berlin. This approach involves harvesting a patient's peripheral blood mononuclear cells and expanding them ex vivo into cytokine-induced killer (CIK) cells—a heterogeneous population of immune effectors with T-cell and NK-cell properties. These cells are then genetically modified via electroporation with a plasmid containing the interleukin-2 (IL-2) gene. The resulting cells produce IL-2 (ranging from 330-1800 pg/10^6 cells/24h), which acts in an autocrine and paracrine manner to enhance their proliferation and tumor-specific cytotoxic activity. Administered intravenously, this therapy has been investigated in Phase I trials for metastatic renal cell carcinoma, metastatic colorectal cancer, and lymphoma, showing a favorable safety profile and potential clinical activity, including a complete response in a lymphoma patient.
