Overview

IL-21 expanded third-party natural killer cells + mogamulizumab is an investigational combination immunotherapy regimen being evaluated for the treatment of relapsed or refractory cutaneous T-cell lymphoma (CTCL) and adult T-cell leukemia/lymphoma (ATLL). The regimen consists of allogeneic, "off-the-shelf" natural killer (NK) cells that have been expanded ex vivo using interleukin-21 (IL-21) to enhance their activation, proliferation, and cytotoxic potential. These cells are administered in conjunction with mogamulizumab, a humanized monoclonal antibody that targets the CC chemokine receptor 4 (CCR4), which is highly expressed on the surface of malignant T-cells. The primary mechanism of action is antibody-dependent cellular cytotoxicity (ADCC), in which the infused NK cells recognize and bind to the Fc portion of mogamulizumab via their CD16 receptors, leading to the targeted lysis of the cancerous T-cells. This combination approach is designed to augment the anti-tumor activity of mogamulizumab by providing a potent, exogenous source of effector cells.
