Interleukin-22 (IL-22) is a cytokine belonging to the interleukin-10 family, produced by various immune cells including T helper 17 (Th17), Th22 cells, and innate lymphoid cells. IL-22 primarily acts on nonhematopoietic cells such as epithelial and stromal cells due to the restricted expression of its receptor complex (IL-22R1/IL-10R2). Its main functions include regulation of epithelial homeostasis, promotion of tissue repair and regeneration, enhancement of antimicrobial peptide production, modulation of barrier function in organs like the intestine and lung, and protection against bacterial infections. Mechanistically, IL-22 signals through activation of Jak1/Tyk2 kinases leading to STAT3 phosphorylation as well as activation of AKT/mTOR, ERK1/2, P38 MAPK pathways. It is being explored for therapeutic potential in diseases involving epithelial damage or inflammation such as inflammatory bowel disease (IBD), graft-versus-host disease (GVHD), lung injury including COVID-related damage, and certain cancers[2][5][4].
