Drug intelligence / Profile preview
IL-233 (also known as IL233) is a preclinical-stage, first-in-class bifunctional fusion cytokine that combines the active domains of interleukin-2 (IL-2) and interleukin-33 (IL-33) into a single hybrid molecule. Developed by Slate Bio (a spinout from the University of Virginia), IL-233 acts as a dual agonist of the IL-2 receptor (IL-2R) and the IL-33 receptor (ST2/IL1RL1). By simultaneously engaging both pathways, IL-233 synergistically promotes the expansion, activation, and tissue-homing of regulatory T cells (Tregs) and group 2 innate lymphoid cells (ILC2s). This dual agonism restores immune homeostasis and suppresses excessive Th1/CD8-driven inflammatory responses. Preclinical studies have demonstrated that IL-233 can prevent or delay acute graft-versus-host disease (aGvHD), induce long-lasting remission in mouse models of lupus glomerulonephritis, and protect against diabetic nephropathy and acute kidney injury.
