Overview

IL-23R shRNA is a gene-silencing therapeutic candidate designed to knock down the expression of the interleukin-23 receptor (IL-23R). Research presented at ASH 2023 revealed that IL-23R is expressed intracellularly in acute myeloid leukemia (AML) cells, where it serves as a critical regulator of the mitotic spindle and centrosome formation. By utilizing short hairpin RNA (shRNA) to target IL-23R mRNA, this approach induces mitotic spindle dysregulation, including multipolarity and lagging chromosomes, which leads to reduced cell proliferation and viability. Preclinical studies have demonstrated that IL-23R knockdown impairs the engraftment of AML cells in murine models while sparing normal hematopoietic cells, highlighting its potential as a selective therapeutic strategy for AML.
