IL-2c is a chimeric interleukin-2 (IL-2) mutein designed to enhance the therapeutic index of IL-2-based cancer immunotherapy. Developed by Anwita Biosciences, IL-2c is engineered by incorporating a fragment from IL-15, resulting in a molecule that retains high affinity for the IL-2 receptor beta and gamma subunits (IL-2Rβγ) but lacks binding to the IL-2 receptor alpha subunit (IL-2Rα/CD25). This "No-alpha" design is intended to selectively activate effector T cells and natural killer (NK) cells while avoiding the stimulation of immunosuppressive regulatory T cells (Tregs) and minimizing systemic toxicities such as vascular leak syndrome. IL-2c is frequently utilized as a functional payload in bifunctional fusion proteins, such as the anti-PD-1-IL-2c fusion (αPD-1-IL-2c), to achieve targeted immune activation and expansion within the tumor microenvironment.
