IL-2cx (Interleukin-2 complex) is an immunotherapy consisting of recombinant human interleukin-2 (IL-2) non-covalently bound to a specific anti-IL-2 monoclonal antibody. This complex is designed to overcome the limitations of recombinant IL-2 therapy, such as its short half-life and the induction of regulatory T cells (Tregs) via the high-affinity IL-2 receptor alpha (CD25). By complexing IL-2 with an antibody that masks the CD25-binding site, IL-2cx selectively directs IL-2 signaling toward the dimeric IL-2 receptor beta/gamma (CD122/CD132) expressed on natural killer (NK) cells and memory CD8+ T cells. This selective activation promotes the de novo generation and massive expansion of functionally mature NK cell precursors. In preclinical studies, IL-2cx has shown efficacy in controlling Acute Myeloid Leukemia (AML) and lymphoma, particularly as a post-transplant immunotherapy to enhance graft-versus-leukemia effects without the need for adoptive cell transfer.
