The IL-31 vaccine is an investigational immunotherapeutic designed to induce an immune response against interleukin 31 (IL-31), a cytokine implicated in the pathogenesis of pruritic (itch-associated) inflammatory skin diseases such as atopic dermatitis and insect bite hypersensitivity. The mechanism of action involves generating antibodies that neutralize IL-31, thereby inhibiting its signaling through the IL-31 receptor complex (composed of IL-31RA and OSMRβ subunits). This blockade reduces pruritus and associated inflammation by preventing downstream activation of JAK/STAT, PI3K/AKT, and MAPK pathways. Preclinical studies have demonstrated efficacy in animal models—such as horses with insect bite hypersensitivity—using vaccines like eIL‑31-CuMVTT (equine interleukin 31 conjugated to a virus-like particle platform) and WISIT vaccines based on human or animal-derived peptides. These vaccines are being developed for chronic pruritic dermatoses including atopic dermatitis[1][2][4][7].
