IL-31RA siRNA is a small interfering RNA (siRNA) designed to silence the expression of the interleukin-31 receptor alpha (IL-31RA) subunit. Research presented at the ATS 2025 conference indicates that IL-31RA plays a critical, ligand-independent role in regulating airway smooth muscle cell (ASMC) contraction and airway hyperresponsiveness (AHR) in asthma. By knocking down IL-31RA in human ASMCs, researchers observed a significant reduction in calcium elevation and myosin light chain phosphorylation in response to contractile stimuli like carbachol and histamine. This effect is mediated by the downregulation of cell surface expression of the M3 muscarinic receptor (CHRM3) and Histamine H1 Receptor (H1HR). These findings suggest that targeting IL-31RA with siRNA or other inhibitors could provide a novel therapeutic approach for managing AHR in asthmatic patients.
