IL-4-Kinoid is an experimental therapeutic vaccine consisting of interleukin-4 (IL-4) chemically conjugated to a carrier protein (typically the non-toxic mutant diphtheria toxin CRM197), forming a so‑called kinoid that breaks self-tolerance and induces polyclonal neutralizing antibodies against IL-4. By eliciting long-lasting endogenous antibodies that neutralize IL-4, the vaccine dampens type 2 helper T cell (Th2) responses, reduces IgE production, eosinophilia, airway hyperresponsiveness, mucus hypersecretion, and mast cell activation in preclinical models of allergic asthma and anaphylaxis.[1][15][4] The kinoid platform and IL-4/IL-13 kinoid program have been developed primarily by the French biotech Neovacs in collaboration with academic partners such as INSERM and Institut Pasteur for long-term treatment of allergic diseases including asthma.[4][10][11]
