Overview

IL-4 transfected fibroblasts refers to a cellular therapy approach in which patient-derived fibroblasts are genetically modified to express interleukin-4 (IL-4), a cytokine, using a retroviral vector (such as TFG-IL4-Neo-TK). The therapeutic concept involves mixing these IL-4-expressing fibroblasts with autologous tumor cells and administering the mixture as a vaccine to stimulate an anti-tumor immune response. This experimental therapy has been investigated in clinical trials for malignant gliomas. The aim is to take advantage of IL-4's ability to enhance dendritic cell maturation (thereby potentiating dendritic cell vaccines) and to attract immune cells, such as CD4+ and CD8+ T cells, to the tumor microenvironment. The therapy incorporates a suicide gene cassette (HSV-TK) for safety, enabling the selective elimination of any persistent live tumor cells after immune response[1].
