Drug intelligence / Profile preview
An IL-4Rα (interleukin-4 receptor alpha) blocking antibody is a class of monoclonal antibodies designed to target the alpha subunit of the IL-4 receptor (IL-4Rα). This subunit is a shared component of both the Type I IL-4 receptor complex (which binds IL-4) and the Type II IL-4 receptor complex (which binds both IL-4 and IL-13). By blocking IL-4Rα, these antibodies inhibit downstream JAK/STAT signaling pathways, thereby suppressing Type 2 helper T cell (Th2)-mediated inflammatory cascades. This therapeutic approach is highly effective and approved for various Type 2 inflammatory diseases, including atopic dermatitis, asthma, and chronic rhinosinusitis with nasal polyposis (e.g., dupilumab). Additionally, emerging preclinical and clinical research suggests that IL-4Rα blockade can reverse immunosuppressive myeloid cell programs (such as monocyte-derived macrophages) within the tumor microenvironment, offering a potential strategy to overcome immunotherapy resistance in cancers like colorectal cancer and non-small cell lung cancer.
