IL-5 CAR-T cell therapy is an experimental chimeric antigen receptor (CAR) T-cell therapy designed to treat refractory eosinophil-driven respiratory and autoimmune diseases. Developed by researchers at Zhejiang University, this therapy utilizes human interleukin-5 (IL-5) as the antigen-binding domain of the CAR to selectively target and eliminate cells expressing the IL-5 receptor alpha (IL-5Rα). This includes mature pathogenic eosinophils and their bone marrow progenitors. By depleting these cells, the therapy aims to provide sustained remission in conditions such as severe asthma, eosinophilic granulomatosis with polyangiitis (EGPA), and hypereosinophilic syndromes (HES). Preclinical studies have demonstrated potent cytotoxicity and tumor regression in eosinophilic leukemia models with a favorable safety profile, and GMP-grade manufacturing processes have been established for clinical translation.
