Overview

IL-6 shRNA (short hairpin RNA targeting interleukin-6) is a gene-silencing therapeutic platform designed to knock down the expression of interleukin-6 (IL-6), a key pro-inflammatory cytokine. By utilizing RNA interference (RNAi), IL-6 shRNA sequences are typically delivered via viral vectors (such as lentivirus or adeno-associated virus) or lipid nanoparticles to target cells. In oncology, IL-6 shRNA has been investigated to suppress autocrine and paracrine IL-6 signaling, which drives tumor growth, metastasis, and drug resistance in cancers like breast cancer, ovarian cancer, and osteosarcoma. Additionally, IL-6 shRNA is integrated into chimeric antigen receptor (CAR) T-cell therapies (creating ssCAR-T cells) to autonomously downregulate IL-6 expression, thereby mitigating cytokine release syndrome (CRS) and immune effector cell-associated neurotoxicity syndrome (ICANS) without compromising antitumor efficacy.
