VRG Therapeutics is developing a miniprotein therapeutic designed to selectively inhibit IL-6 trans-signaling for the treatment of chronic inflammatory and autoimmune diseases. Developed using the company's AI-powered miniprotein platform (AI-MPRO) and WISDOM technology, this candidate aims to block the pro-inflammatory trans-signaling pathway (mediated by the soluble IL-6 receptor) while preserving the homeostatic classical signaling pathway (mediated by the membrane-bound IL-6 receptor). This selectivity is intended to reduce the side effects associated with global IL-6 inhibition, such as increased infection risk and neutropenia. Miniproteins are small, disulfide-stabilized peptides that combine the high affinity and specificity of monoclonal antibodies with the stability and potential for oral delivery characteristic of small molecules. The program is currently in the early research stage.
