IL-7-armed adenovirus is an engineered oncolytic adenovirus (OAd) designed to express the cytokine interleukin-7 (IL-7) within the tumor microenvironment. Developed as a viro-immunotherapy, it combines the direct oncolytic activity of the adenovirus—which selectively replicates in and lyses cancer cells—with the immunomodulatory properties of IL-7. IL-7 is a critical cytokine for the development, survival, and homeostatic proliferation of T-cells. By delivering IL-7 directly to the tumor, the therapy aims to overcome the immunosuppressive tumor microenvironment (TME), enhance the infiltration and persistence of anti-tumor immune cells, and potentially synergize with adoptive T-cell therapies such as CAR T-cells. Preclinical research, notably at the University of Minnesota, has explored its efficacy in models of pancreatic ductal adenocarcinoma (PDAC), demonstrating significant tumor growth inhibition, reduction in fibrosis, and abscopal effects on distant metastatic lesions.
