An IL-7R blocking antibody is a monoclonal antibody designed to target and inhibit the Interleukin-7 receptor (IL-7R), specifically the alpha chain known as CD127. By binding to CD127, these antibodies prevent the binding of IL-7, thereby inhibiting the activation of the JAK/STAT5 signaling pathway. This pathway is critical for the survival, development, and homeostatic proliferation of T cells and certain B-cell progenitors. Dysregulation of IL-7/IL-7R signaling is implicated in various autoimmune diseases, such as ulcerative colitis and multiple sclerosis, as well as in lymphoid malignancies like T-cell acute lymphoblastic leukemia (T-ALL). In the context of B-cell acute lymphoblastic leukemia (B-ALL), IL-7R blockade is being researched for its potential to influence lineage stability and prevent lineage switching to acute myeloid leukemia (AML).
