IL10-TCE is an experimental, next-generation T-cell engager (TCE) designed to treat solid tumors by addressing T-cell dysfunction in the tumor microenvironment. It consists of an IL-10 cytokine fused to the N-terminus of an anti-CD3 arm via a cleavable linker, a configuration referred to as a "pro-TCE." This design aims to deliver IL-10 specifically to the tumor site, where IL-10 receptors are enriched on antigen-specific T cells. By providing localized IL-10 signaling, the drug counteracts activation-induced cell death (AICD) and T-cell exhaustion, which are common limitations of traditional TCE therapies. Preclinical studies presented at the AACR 2026 Annual Meeting demonstrated that IL10-TCE promotes the expansion of tumor-infiltrating lymphocytes and leads to the complete regression of established solid tumors and metastatic lesions in models of colon cancer, melanoma, and pancreatic cancer, without systemic toxicity.
