IL12-MSLN CAR NK cells are genetically engineered natural killer (NK) cells designed for cancer immunotherapy. These cells are modified to express a chimeric antigen receptor (CAR) targeting mesothelin (MSLN), a membrane glycoprotein overexpressed on various solid tumors, notably in ovarian, pancreatic, and certain hematologic cancers. In addition, they are engineered to secrete interleukin-12 (IL-12), a cytokine known to enhance anti-tumor immune responses. The combination improves the NK cells' activation, metabolic fitness, cytotoxicity, and cytokine production. IL12-MSLN CAR NK cells have demonstrated increased killing of MSLN-positive cancer cells, higher IFN-γ production, and greater induction of MHC I/II on target tumor cells compared to MSLN CAR NK cells lacking IL-12. Preclinical studies in acute myeloid leukemia (AML) and solid tumors have shown promising efficacy, supporting their further evaluation in relapsed/refractory MSLN-positive malignancies[1][2][3].
