IL12-XPAC is a protease-activated, masked interleukin‑12 (IL‑12) cytokine therapy being developed using Amunix’s XPAC platform, which is designed to create long-acting, conditionally activated biologics that remain inert systemically and are preferentially unmasked by tumor-associated proteases in the tumor microenvironment.[7][11][13][14][15][16] By combining IL‑12’s potent immunostimulatory activity with tumor-restricted activation, IL12-XPAC aims to enhance antitumor T‑cell and NK‑cell responses while limiting the systemic cytokine-mediated toxicities that have constrained systemic IL‑12 therapies in prior clinical development.[1][3][4][5][7][13][14][16] The program is currently in the discovery/preclinical stage within the oncology portfolio originally advanced by Amunix and now part of Sanofi’s oncology innovation efforts following its acquisition of Amunix.[7][9][11][13][14][15][16]
