Overview

IL13Rα2-targeted CAR-T cells are an investigational cellular immunotherapy designed to target interleukin-13 receptor alpha 2 (IL13Rα2), a tumor-associated antigen frequently overexpressed in high-grade gliomas, including glioblastoma multiforme (GBM), while showing limited expression in normal brain tissue. The therapy involves engineering a patient's T cells to express a chimeric antigen receptor (CAR) that recognizes IL13Rα2, typically using a mutated IL-13 cytokine ligand (e.g., IL-13 E13Y) or a single-chain variable fragment (scFv) as the binding domain. These CAR-T cells are often administered via intracranial (intratumoral or intraventricular) or intravenous routes. Clinical development is being pursued by several institutions and companies, including City of Hope (MB-101) and Cellabmed (CLM-101), focusing on recurrent malignant gliomas and other primary brain tumors.
