IL13Rα2 x CD3 DART is a bispecific Dual-Affinity Re-Targeting (DART) protein developed by MacroGenics for the treatment of solid tumors expressing the interleukin-13 receptor alpha 2 (IL13Rα2). The molecule is designed to simultaneously engage IL13Rα2 on tumor cells and the CD3 epsilon chain on cytotoxic T cells, thereby facilitating T-cell activation and redirected killing of the target tumor cells. The construct incorporates an Fc domain to extend its circulating half-life. Preclinical studies have demonstrated potent anti-tumor activity and cytokine release in models of glioblastoma, melanoma, lung adenocarcinoma, and medulloblastoma. While it was identified as a development candidate, it does not currently appear in MacroGenics' active clinical pipeline.
