Overview

IL13Ralpha2 hinge-optimized 41BB CAR truncated CD19-expressing autologous T-cells is a cellular immunotherapy that uses a patient's own T cells, which are genetically modified to express a chimeric antigen receptor (CAR) targeting IL13Rα2, a protein frequently overexpressed in glioblastoma and other malignant brain tumors. The therapy incorporates a 4-1BB (CD137) co-stimulatory domain for improved T cell persistence and anti-tumor activity, along with a truncated CD19 marker for cell tracking. The CAR has been designed with an optimized hinge region to enhance binding to IL13Rα2 on tumor cells while minimizing binding to IL13Rα1, which is more widely expressed in normal tissues. This therapy is administered locoregionally (intratumoral and/or intraventricular) to patients with recurrent or refractory malignant brain tumors.
