Overview

IL1RAP CAR-T is an investigational chimeric antigen receptor (CAR) T-cell therapy targeting the interleukin-1 receptor accessory protein (IL1RAP). IL1RAP is highly expressed on acute myeloid leukemia (AML) blasts and leukemic stem cells but is absent on normal hematopoietic stem cells, making it a promising target for selective AML therapy. Developed by researchers at City of Hope, this therapy aims to overcome challenges in AML treatment such as antigen heterogeneity and a suppressive tumor microenvironment. Recent research has explored the use of microRNA-142 (miR-142) to improve the metabolic fitness, persistence, and antileukemic activity of these CAR-T cells by reducing apoptosis and enhancing glycolysis and oxidative phosphorylation.
