Overview

IL1RAP-mut CAR-T is a chimeric antigen receptor (CAR) T-cell construct featuring a mutated version of the interleukin-1 receptor accessory protein (IL1RAP) binding domain. It was developed by researchers at the City of Hope National Medical Center as a negative control for IL1RAP-targeting CAR-T therapies in the treatment of acute myeloid leukemia (AML). Unlike the functional IL1RAP CAR-T, which targets IL1RAP-expressing AML blasts and leukemic stem cells, the mutated version lacks specific antileukemic activity, serving to validate that the therapeutic effects of the wild-type CAR-T are mediated through specific antigen recognition.
