IL2401 is a small-molecule targeted protein degrader (TPD) developed by iLeadBMS for the treatment of various solid tumors. It specifically targets Cyclin-K (CCNK), a regulatory protein that interacts with cyclin-dependent kinases such as CDK12 and CDK13 to modulate RNA polymerase II-mediated transcription and the DNA damage response. By inducing the selective degradation of Cyclin-K, IL2401 disrupts transcriptional elongation and cellular homeostasis, leading to potent anti-tumor activity. The compound is currently being investigated for its potential in treating gastric cancer, breast cancer, colorectal cancer, and triple-negative breast cancer (TNBC).
