IL27-expressing oncolytic adenovirus (oAd-IL27) is an investigational oncolytic virus therapy engineered to treat advanced prostate cancer. The therapy utilizes a genetically modified adenovirus that selectively replicates within and lyses cancer cells (oncolysis) while simultaneously serving as a gene delivery vehicle for Interleukin 27 (IL-27). IL-27 is a cytokine known to promote anti-tumor immune responses and suppress tumor growth and metastasis without the systemic toxicity typically associated with cytokine storms. To protect the virus from pre-existing neutralizing antibodies and enhance delivery to metastatic sites, researchers have explored using adipose-derived mesenchymal stromal cells (ASCs) as a delivery vehicle. This approach aims to combine direct viral-mediated cell death with localized, sustained cytokine-driven immunotherapy to modulate the tumor microenvironment.
