IL33 siRNA refers to a class of small interfering RNA (siRNA) molecules designed to silence the expression of Interleukin-33 (IL33), a key alarmin cytokine involved in the initiation and maintenance of type-2, type-1, and regulatory immune responses. IL33 is highly expressed in alveolar type 2 (AT2) and endothelial cells in the lung and is a significant susceptibility locus for asthma. By utilizing RNA interference (RNAi), these molecules target and degrade IL33 mRNA, leading to a durable reduction in IL33 protein levels. Arrowhead Pharmaceuticals is developing lung-targeted IL33 siRNAs, including human cross-reactive versions and dual-silencing dimer platforms (targeting both IL33 and TSLP), for the treatment of chronic inflammatory lung diseases such as asthma and chronic obstructive pulmonary disease (COPD). These therapies are typically designed for inhaled delivery to provide localized, potent, and long-lasting anti-inflammatory effects.
