IL4RPep-1-Abx (also known as IL4R-Abx) is an interleukin-4 receptor (IL4R)-targeted nanoparticle conjugate. It is constructed by conjugating the IL4R-binding peptide IL4RPep-1 to Abraxane (nab-paclitaxel, an albumin-bound paclitaxel nanoparticle) using click chemistry. IL4R is highly expressed on certain tumor cells and pro-tumoral M2-polarized tumor-associated macrophages (TAMs). By targeting IL4R, IL4RPep-1-Abx enhances the delivery of paclitaxel to the tumor microenvironment, induces cytotoxicity in IL4R-expressing cancer cells, and promotes the reprogramming of immunosuppressive M2 macrophages into immunostimulatory M1-like phenotypes via the ROS-HMGB1-TLR4 pathway. This dual action enhances antitumor immunity and inhibits tumor growth and metastasis.
