IL4RPep-1-labeled cytotoxic T cells are a non-genetically engineered adoptive cell therapy consisting of cytotoxic T lymphocytes (CTLs) surface-modified with the IL-4 receptor (IL-4R)-binding peptide IL4RPep-1 (sequence: CRKRLDRNC). The peptide is anchored to the T cell membrane using dioleylphosphatidylethanolamine (DOPE) via a biological anchor for membrane (BAM) linker. This modification enhances the homing and binding of CTLs to IL-4R-overexpressing tumor cells, such as melanoma. In preclinical models, these targeted CTLs demonstrate increased tumor infiltration, enhanced cytokine secretion (e.g., interferon-gamma), and the ability to reprogram the tumor microenvironment by shifting immunosuppressive M2 macrophages toward an anti-tumor M1 phenotype. This research-stage modality was developed by researchers at Kyungpook National University.
