**IL7Fc-armored PSMA CAR T-cells** is a preclinical family of engineered, PSMA-directed CAR-T-cell candidates developed by Lentigen Technology. The cells express a chimeric antigen receptor directed against prostate-specific membrane antigen and are armored to secrete a human interleukin-7-human IgG1 Fc fusion protein intended to improve CAR-T-cell proliferation, stem-like memory phenotype, and persistence in prostate cancer. The reported program includes PSMA2IL7Fc and PSMA14IL7Fc constructs; although armoring improved in-vitro expansion and serial tumor-cell killing, high systemic human IL-7, IFN-gamma, and GM-CSF were associated with lethal toxicity in an LNCaP xenograft mouse study, supporting the need for controlled or inducible IL7Fc expression.
