IL8 shRNA is an experimental RNA interference (RNAi) therapeutic designed to silence the expression of Interleukin-8 (IL8/CXCL8). IL8 is a pro-inflammatory chemokine that plays a critical role in the tumor microenvironment by promoting the proliferation, migration, and survival of cancer cells, particularly colon cancer-initiating cells (CCICs). By delivering short hairpin RNA sequences that target IL8 mRNA for degradation, this approach aims to inhibit the autocrine and paracrine signaling pathways that drive tumorigenicity and the progression from chronic inflammation (such as ulcerative colitis) to colitis-associated cancer. Preclinical studies have demonstrated that IL8 knockdown significantly reduces tumor growth in xenograft models.
