Ilepcimide (also known as Antiepilepsirine) is an antiepileptic drug that has been used clinically in China for decades. Its antiepileptic effect is primarily attributed to the inhibition of voltage-gated sodium channel activity in hippocampal neurons. Beyond its established use in epilepsy, ilepcimide has shown potential for repurposing in Multiple Sclerosis (MS), where it ameliorates experimental autoimmune encephalomyelitis (EAE), a mouse model of MS, by restricting inflammatory responses and oxidative stress. This protective effect in MS models involves inhibiting dihydroorotate dehydrogenase (DHODH) and activating the NRF2 antioxidant pathway. It is structurally similar to the antiepileptic drug lamotrigine and is an artificial synthetic piperine analog.
