Drug intelligence / Profile preview
ILF2-ASO is an antisense oligonucleotide (ASO) designed with constrained ethyl (cEt) chemistry to target and deplete interleukin enhancer-binding factor 2 (ILF2). Developed through a collaboration between Ionis Pharmaceuticals and researchers at the University of Texas MD Anderson Cancer Center, ILF2-ASO is being investigated for the treatment of 1q21-amplified multiple myeloma (MM). ILF2 is an RNA-binding protein and a key modulator of the DNA repair pathway that is overexpressed in approximately 70% of patients with relapsed multiple myeloma, promoting tolerance to genomic instability and resistance to DNA-damaging agents. By depleting ILF2, ILF2-ASO induces DNA damage activation, inhibits cell proliferation, and sensitizes myeloma cells to genotoxic therapies such as melphalan, bortezomib, and olaparib.
