This preclinical-stage small molecule program is being developed by Ilios Therapeutics for the treatment of Amyotrophic Lateral Sclerosis (ALS) and Frontotemporal Dementia (FTD). The lead program focuses on multi-functional, brain-penetrant small molecules designed to address three primary pathological drivers of neurodegeneration: correcting TAR DNA-binding protein 43 (TDP-43) mislocalization, modulating Nuclear factor erythroid 2-related factor 2 (NRF2) dysregulation, and reducing microglial inflammation. The therapeutic approach utilizes a proprietary platform that conjugates bioactive pharmacophores with glutathione and fatty acids to enhance penetration across the blood-brain barrier. The program is currently in the lead generation and preclinical development phase.
